NSBP1 binds specifically to nuclear core particles and acts as a transcriptional activator. [Subcellular Location] Nucleus. [Tissue Specificity] Widely expressed with highest levels in submaxillary gland, thymus, kidney and liver and lowest levels in brain, lung, pancreas and eye. [Developmental Stage] Highest levels are found in 7-day-old embryos. Levels in the 7-day-old embryo are 4-fold higher than in the adult and almost 10-fold higher than in later embryonic stages. [SIMILARITY] Belongs to the HMGN family. Function: Preferentially binds to euchromatin and modulates cellular transcription by counteracting linker histone-mediated chromatin compaction (By similarity). Subcellular Location: Nucleus. Note=Associates with nucleosomes in euchromatin and is largely excluded from constitutive heterochromatin (By similarity). Tissue Specificity: Ubiquitously expressed. Paraformaldehyde-fixed, paraffin embedded (human testis); Antigen retrieval by boiling in sodium citrate buffer (pH6.0) for 15min; Block endogenous peroxidase by 3% hydrogen peroxide for 20 minutes; Blocking buffer (normal goat serum) at 37°C for 30min; Antibody incubation with (NSBP1 ) Polyclonal Antibody, Unconjugated (bs-0700R) at 1:200 overnight at 4°C, followed by operating according to SP Kit(Rabbit) (sp-0023) instructionsand DAB staining.
Tissue/cell: mouse embryo tissue; 4% Paraformaldehyde-fixed and paraffin-embedded;
Antigen retrieval: citrate buffer ( 0.01M, pH 6.0 ), Boiling bathing for 15min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30min; Blocking buffer (normal goat serum,C-0005) at 37℃ for 20 min;
Incubation: Anti-NSBP1 Polyclonal Antibody, Unconjugated(bs-0700R) 1:200, overnight at 4°C, followed by conjugation to the secondary antibody(SP-0023) and DAB(C-0010) staining
